"Glee" star Jane Lynch is divorcing her wife of three years.

Lynch married Dr. Lara Embry in 2010 in Massachusetts. She told People magazine in a statement Monday that splitting up was "a difficult decision for us as we care very deeply about one another."

Lynch asked for privacy for the family, which includes Embry's daughter.

The 52-year-old actress met Embry, a 44-year-old clinical psychologist, in 2009 and they wed on Memorial Day in 2010.

Lynch is currently starring in "Annie" on Broadway and performed on Sunday night's Tony Awards on CBS.

Epigenetic factor likely plays a key role in fueling most common childhood cancer

June 10, 2013 ? Changes in an epigenetic mechanism that turns expression of genes on and off may be as important as genetic alterations in causing pediatric acute lymphoblastic leukemia (ALL), according to a study led by scientists at St. Jude Children's Research Hospital and published in the June 10 online edition of the Journal of Clinical Investigation.

The results suggest the mechanism called cytosine methylation plays a previously under-appreciated role in the development of leukemia. Cytosine methylation involves adding or removing methyl groups to cytosine, which is a building block of DNA.

The study is the most comprehensive effort yet to identify and understand genetic and epigenetic factors that work together to cause ALL, the most common childhood cancer. ALL is a cancer of white blood cells known as lymphocytes. Scientists at St. Jude and Weill Cornell Medical College collaborated on the project.

Researchers used a variety of techniques to examine hundreds of thousands of methylation sites across the genome in normal and leukemic lymphocytes, including samples from more than 160 children with ALL. Investigators found that known ALL subgroups, which are defined by chromosomal alterations, have unique methylation profiles. Those profiles correlated with different patterns of gene expression.

"It is well known that different leukemia subgroups have distinct patterns of gene expression that are important in the development of leukemia," said Charles Mullighan, MBBS (Hons), MSc, M.D., an associate member of the St. Jude Department of Pathology. Mullighan and Ari Melnick, M.D., Gebroe Professor Hematology/Oncology at Weill Cornell Medical College, are the study's co-corresponding authors.

"We have assumed that the underlying genetic changes are important determinants of those gene expression profiles. We now know that changes in methylation state also have key roles in influencing gene expression," Mullighan said.

The study used tissue samples from 137 St. Jude patients with B-cell leukemia and 30 children with T-cell leukemia. The patients represented all major ALL subgroups.

"The data show that aberrant epigenetic gene programming can now be considered a hallmark of acute lymphoblastic leukemia, occurring in all patients regardless of the presence of genetic mutations," Melnick said. "This offers the opportunity for development of epigenetic targeted therapies for patients with ALL that could be broadly applicable to many patients."

For comparison, researchers also checked B and T cells from 27 healthy children. Investigators found that leukemia cells shared a core group of abnormally methylated genes. The genes included ones involved in regulating the cell division and proliferation. "This remains to be tested, but the findings suggest that alterations in methylation are an important early step in the development of leukemia," Mullighan said.

The research provides further evidence that genetic and epigenetic events are both important in establishing different subgroups of ALL. For this study, researchers conducted genome-wide sampling of methylation, gene expression and DNA structural abnormalities, including the gain or loss of DNA. Shann-Ching Chen, Ph.D., St. Jude Pathology, developed many of the methods used to integrate and analyze the results. Chen and Maria Figueroa, now of the University of Michigan and formerly of Cornell, are the study's co-first authors.

The study also found that more than one-third of 71 genes targeted by genetic alterations are also abnormally methylated in ALL. The methylation changes involved known tumor suppressor or oncogenes genes including CDKN2A, CDKN2B, PTEN and KRAS. "The findings suggest these genes are inactivated or deregulated more frequently than suggested by simply analyzing structural changes in the genome," Mullighan said.

The other authors are Anna Andersson, formerly of St. Jude and now of Lund University Hospital, Sweden; Letha Phillips, formerly of St. Jude; Mondira Kundu and James Downing, both of St. Jude; Yushan Li, Cornell; and Jason Sotzen, University of Michigan.

Many Game of Thrones fans worry that George R.R. Martin may not be able to finish the remaining two books in his A Song of Ice and Fire series fast enough to keep up with the HBO series. In an appearance on Conan, Martin said the HBO show is ?moving like a freight train.? But maybe there?s nothing to worry about. The above chart uses the average time between Song of Ice and Fire book releases to predict the times Martin will complete the remaining two books and compares this with the expected schedule of the show, which is supposed to run for eight seasons. (The third book was split into two seasons.) If Martin writes as fast as he normally does, he will have a little time to spare. It?ll be close, but he?ll make it.

This, of course, assumes Martin doesn?t take an exceptionally long time with the final two books?and he might. On Conan he described The Winds of Winter as a ?fifteen-hundred page monster,? which would make it as long as A Dance With Dragons. If the last two take the six years that Martin took to release A Dance with Dragons, then expect the HBO show to go on hiatus or produce an ending based on hints from Martin. And who knows that there won?t ultimately be eight or more books? The series was originally supposed to be a trilogy.

Still, there are bigger problems to worry about?for example, hiatus or no hiatus, the teenage actors who play Arya, Bran, and Sansa are growing up much faster than their characters. Then again, maybe their characters will die in horrendous fashion before that becomes too noticeable.

A precocious moppet with an edge will face off against a proud drag queen when the annual Tony Awards, Broadway's highest honors, are bestowed on Sunday night.

"Kinky Boots," an adaptation of a British film about a struggling shoe factory reinventing itself by making boots for drag queens, topped the Tony nominations with 13.

But many Broadway experts see the British import "Matilda," which boasts ecstatic reviews and 12 nods, prevailing as best new musical.

"This is a two-way race," said Entertainment Weekly's Thom Geier about the awards.

The "Boots" team includes Broadway veterans Harvey Fierstein, director Jerry Mitchell and music by pop star Cyndi Lauper. Its story bears the hallmarks of acceptance and triumph, which would likely play well on tours of the heartland.

"Matilda," based on a story by Roald Dahl about a freakishly intelligent little girl's persecution by her crass family and a fearsome school headmistress, has collected the lion's share of early prizes including the Drama Desk.

But following the Tony nominations its box office receipts dropped slightly, while the take for "Kinky Boots" edged up.

Goldderby.com, a website that tracks show business awards, polled 14 Broadway experts and found a near split for the top prize, with a slight edge going to "Matilda."

"The production made history at the Olivier Awards, winning a record seven of ten bids," the website said, adding "Kinky Boots" nabbed some early best musical honors "and could pull off an upset."

Patrick Healy of the New York Times predicted "the warmth of 'Kinky Boots' will be enough to score an upset."

Clear Favorites

On the drama side most experts predict the best actor award will go to Hollywood star Tom Hanks for his Broadway debut in the late Nora Ephron's journalism drama, "Lucky Guy."

They also see veteran playwright Christopher Durang winning his first best play Tony for his crowd-pleasing comic riff on Chekhov, "Vanya and Sonia and Masha and Spike," which stars Sigourney Weaver and Tony nominee David Hyde Pierce.

"The clear front-runner is 'Vanya and Sonia and Masha and Spike,' goldderby.com said, noting "reviews were stellar across-the-board."

Cicily Tyson, marking her return to Broadway after a 30-year absence, is expected to take home the prize for best actress for her moving turn in a revival of "The Trip to Bountiful," while an acclaimed, though shuttered production of "Who's Afraid of Virginia Woolf" is the likely winner for best revival of a play.

A much-praised reworking of the seminal 1970s musical "Pippin" is the clear favorite to win for best musical revival, while the best actor prize is expected to come down to two men wearing dresses.

Billy Porter holds the center of "Kinky Boots" as the noble drag queen, but Bertie Carvel steals the show as the dementedly outrageous school headmistress in "Matilda." All bets are on Patina Miller to win best actress as the host in "Pippin," a part that won Ben Vereen a Tony in the original production.

Whatever the outcome on Sunday, Broadway will emerge victorious. Box office receipts typically spike in the weeks after the awards.

"Year after year we find that shows that win a Tony award do better at the box office," said Charlotte St. Martin, executive director of industry group The Broadway League.

"Once," a small Irish love story with no big name stars, is still going strong a year after sweeping last year's awards, including best musical.

On the flip side, the awards can be irrelevant to a show's fortunes. This season's biggest hit, Berry Gordy's "Motown: The Musical" was snubbed in the best musical category in favor of two limited run musicals, garnering only one major nomination.
